Journal Club This Tuesday

    1. I learned a new method for imaging intracellular proteins at nano-scale resolution using the PALM technique, which produces super-resolved images based on the sum of Gaussian of the standard deviation of PSFs over many images, rather than the deviation of the PSF itself. The process involves the repeated activation, measurement, and bleaching of the fluorescent molecules
    2. This process took up to 12 hours to obtain the image. Is there any method that yields the same resolution in less time?

    4. How does this paper relate to SyBBURE research? Is anyone doing microscopy at this level?

    2.How do PA-FPs work? Is it that they don't lose their fluorescent abilities and you just "recharge" them with light? How did the inactivated, unbleached molecules get depleted?

    Why/how is this method better than transmission electron microscopy?

    2. So pessimist stephen asks why are they certain the proteins imaged don't simply occupy similar thermodynamically favorable spaces in the focal planes used? or are they sure it not just statistically averaging places proteins are likely to bind since they might have a conformation that favors binding a protein that is not able to be imaged via fluorescence?
